Résumé

Cryopreservation in the cell-adhesion state was investigated for the development of useful and easier handling technique of cell culture. In the present study, we used dimethyl sulfoxide (DMSO) as a cryoprotective agent and determined the suitable DMSO concentration for cryopreservation of HepG2 cells in the cell-adhesion state attached to glass and polydimethylsiloxane (PDMS) matrices coated with poly-Llysine. As a result, the rate of cell adhesion to PDMS matrix remained constant irrespective of the DMSO concentration between 10 v/v% and 30 v/v%. In the case of glass matrix, on the other hand, the rate of cell adhesion increased with the DMSO concentration. Cell viability was the highest at 20 v/v% DMSO concentration. DMSO concentration of 30 v/v%, however, had a cytotoxic effect on cell viability. Thus, 20 v/v% DMSO concentration was found to be the most suitable for cryopreservation of HepG2 cells in the adhesion state.
