Résumé

In order to clarify the cryoprotective effect of carboxylated poly-L-lysine (PLL), we investigated freezing behavior and freezing survival of the nematode Caenorhabditis elegans in the presence of PLL. In the absence of PLL, all the nematodes became frozen and did not survive. However, some nematodes remained unfrozen in 20% PLL solution, and 26% of the total nematodes survived after thawing. The synergistic effect of PLL and dimethyl sulfoxide (DMSO) or ethylene glycol (EG) were then investigated, where 5% PLL was dissolved in M9 buffer containing 5% DMSO or 5% EG. Survival rates after freeze-thawing of the nematode in 5% DMSO or 5% EG solution were 82% or 69%, respectively. By the addition of both PLL and DMSO or EG, on the other hand, survival rates were increased to 87% and 74%, respectively.
