Résumé

Genetically modified animals are unique mutants with an enormous scientific potential and rapidly increasing numbers. Cryopreservation of pre-implantation embryos or of spermatozoa and their unlimited storage at -196°C is a valuable alternative to breed those lines. A repository was established with 220.000 cryopreserved embryos from 500 transgenic mouse lines. 300 lines were archived as spermatozoa. The developmental capacity of embryos was demonstrated by using in vitro-cultures and by embryo-transfer of revitalized specimen. Spermatozoa: The in vitro-fertilisation capacity of each line was demonstrated, the quality of each donor assessed by a fluorescence microscopy based, animal-free technique analyzing the membrane integrity. Samples of a line were stored in the liquid phase of LN2 in different permanent freezers at several locations. Possible contaminations during the whole process were investigated. Microorganisms were only found in the freezers. Unexpectedly, dangerous mouse pathogens were detected infrequently following embryo-transfer. A powerful data management system was developed.
