Résumé

Date palm somatic embryos and pea zygotic embryos were pre-treated with glycerol and sucrose cryoprotectant solutions, then dried to specific water content ranges prior to rapid freezing in liquid nitrogen. The addition of low levels of calcium and magnesium to the cryoprotectant solutions reduced the number of embryos regenerating as callus and concomitantly increased the number of embryos differentiating into plantlets. In the case of the pea embryos these additives also increased the total number of axes recovering from cryostorage. The reduction in the callus formation is proposed to be a result of the stabilisation of the subcellular cytoskeletal system by the added elements.
