Résumé

Although cryopreservation of immature oocytes has many benefits for both female fertility preservation and biodiversity conservation, methods still need improvement. The aim of this study was to elucidate which cryobiological changes could affect maturation of bovine cumulus-oocyte complexes (COCs)
Immature bovine COCs were randomly divided into two groups: 1- fresh immCOCs and 2 - cryopreserved immCOCs by vitrification. The number of transzonal projection (TZP) and RNA expression of connexins (Cx) 37, 43, DNMT3a, BAX, BCL2 were evaluated in study groups after in vitro maturation (IVM). These results were compared with maturation, and blastocyst development rates. Our findings showed that cryopreservation decreases the number of TZP and induces significant changes in expression of gap-junction proteins. It also suppresses epigenetic changes normally activated during IVM in fresh COCs. Taken together, these changes led to a dramatic decrease in the rate of IVM and the development of the embryo. Thus, future improvement of the cryopreservation protocol for immCOCs should focused on restoring of the connections between the CCs and oocytes.
