Résumé

Cryopreservation of tissue constructs needs special equipment to ensure reproducible thermal conditions during the whole freezing and thawing process. Standard cell culture vessels are not suited. For that reason special plastic closable dishes were developed and tested in comparison to metallic vessels of comparable measures. Temperature profiles were recorded for several experimental set-ups. That is why a cryoprotocol had to be developed for these experiments. The influence of material on the design of the dishes and the thermal managing tool was demonstrated. Based on tests with Cu-plates which have small heat exchangers a special heat transferring unit was developed. The dishes can be fixed between the two parts of the thermal managing tool (rack) at the beginning of the cryopreservation process and they can stay within this tool during freezing, storage and thawing. This unique tool is applicable in the most freezing units and storage systems.
