Résumé

Currently, freeze-drying is one of the best methods for the preservation of red blood cells. The effects of intracellular trehalose on the viability of freeze-dried red blood cells were investigated. The fresh red blood cells were incubated in trehalose solutions of various concentrations at 37°C for 7 hours. The results show that the intracellular trehalose concentration increased with the increasing concentration of extracellular trehalose solution. The maximum concentration of intracellular trehalose was 35 mmol/L in the study. Red blood cells loaded with trehalose were lyophilized with the concentrations of trehalose of 15, 22.5, 30 and 37.5% respectively. The results show that the viability of freeze-dried red blood cells increased with the increase of intracellular trehalose concentration.
