Résumé

Cryogenic electron microscopy (cryo-EM) is an indispensable tool for studying life sciences and soft matter, enabling the observation of samples in their near-native state under the high vacuum conditions of an electron microscope. In this paper we provide an introduction to cryo-EM and showcase selected applications, ranging from imaging biological macromolecules to investigating soft matter systems, with examples illustrating its transformative impact on these fields. Key methods for sample preparation, such as plunge freezing and high-pressure freezing, will be discussed, along with the potential artefacts and challenges these techniques may introduce. Beyond nanoscale imaging, cryo-EM also facilitates the study of larger, complex structures that are critical for understanding biological and material systems. Aims of this paper is to foster collaboration and mutual understanding between professionals in cryogenics and microscopy.
