Résumé

This review describes the mechanism of freeze-drying preservation of biomaterials by trehalose. Firstly, the vitrification property of this sugar is discussed in comparison with those of other oligosaccharides. Secondly, the phase transition behaviour is discussed on the basis of the results of the author's recent FTIR study, which indicated that the anhydrous form II of trehalose is easily converted to the dehydrated form by adsorption of water. Thirdly, the so-called water replacement hypothesis is discussed on the basis of recent in vitro and in vivo studies, including the author's report on the role of intracellular trehalose in the resistance of yeast cells to water stress.
