Résumé

Different procedures were tested in an attempt to cryopreserve transformed cells of Papaver somniferum in liquid nitrogen. The encapsulation-dehydration process yielded negative results. The vitrification method also proved ineffective. In contrast, using the conventional procedure, suspension cultures resisted to -196 deg C, when treated with 0.75 M sorbitol or maltose and 5% DMSO, and two-step frozen. Better results after freezing were achieved using an original method including encapsulation of cells and conventional cryopreservation. This procedure based on trapping of cells in calcium-alginate allowed easier handling, faster regrowth than for non-encapsulated cells, and sucessful cryopreservation at -196 deg C of Papaver suspensions after a cryoprotective treatment combined with progressive freezing.
