Résumé

Cells of Helianthus tuberius have been successfully recovered after controlled rate freezing, but post-thaw growth was dependent upon the preculture treatment. Mannitol preculture treatments (24 h culture in medium containing 0.5 or 0.75 M mannitol prior to freezing) resulted in significant reductions in prefreeze cell growth and cell viability, as determined by triphenyl tetrazolium chloride (TTC) reduction. No post-thaw cell growth occurred after mannitol preculture treatments. However, the use of sucrose at the same concentrations during preculture, resulted in greater prefreeze cell growth, TTC values and post-thaw cell growth. After preculture with 0.5 M sucrose, sufficient cell growth occurred by 28 days after thawing to allow re-establishment of suspension cultures.
