Résumé

Cold acclimation and freezing injury in plant tissues and isolated protoplasts are described. In addition to molecular, biochemical and biophysical analyses on the freezing tolerance of plants, observation of freezing process under a cryomicroscope seemed to be a powerful tool to understand what happens during freezing and thawing. Cold acclimation resulted in a dramatic effect on the incidence of several freeze-induced lesions associated with the plasma membrane. The differential occurrence of the freeze-induced lesions between non- and cold-acclimated cells was affected by biochemical composition of the plasma membrane. Although further detailed experiments are necessary to understand the complex nature of freezing tolerance, these studies demonstrated that videomicroscopy combined with a cryostage helps us to understand the effect of cold acclimation on the freezing process of plants.
