Potato tubers of high sugar and low sugar accumulating cultivars were stored at 3 deg C for three months. Mitochondria were isolated and their proteins were analyzed. A 26-kiloDa polypeptide, detected only in high sugar accumulating cultivars, was isolated and split into oligopeptides. The fragments were resolved by high-pressure low-chromatography. Partial amino-terminal sequences of two of the oligopeptides were obtained. The corresponding DNA oligomers were synthesized and used as primers in polymerase chain reactions with mitochondrial DNA from high sugar and low sugar selections. One of the amplified sequences was unique to the low sugar selections suggesting a difference in the mitochondrial DNA structure between high sugar and low sugar potato genotypes.
