A novel method for freeze-drying rate measurement.

Résumé

Freezing rate and sublimation rate are both significant parameters during freeze-drying processes, affecting the quality of freeze-dried products and the efficiency of freeze-drying. Traditionally, the freezing rate is obtained through temperature curve and sublimation rate is measured by weighing the samples. These two methods will bring interference to the processes. Image analysis is proposed to investigate the dynamics of freeze-drying process. Pictures of freezing or sublimating samples were taken at intervals by a camera which was fixed in the vacuum cavity together with vials containing samples. Experiments were conducted with mixed solutions of two typical cryoprotective agents: mannitol and sucrose. Very little bio-dyes were added to make the interfaces between frozen part and unfrozen part, between dried part and frozen part more distinct. Through image analysis, the interfaces were determined. The new method can be used along with traditional methods, giving more details of the freeze-drying process.
