Résumé

The purpose of this work is to prepare proliposomes by freeze-drying solution including tertiary butyl alcohol (TBA). Differential scanning calorimetry (DSC) is employed to determine the glass transition temperature of the maximally freeze-concentrated solution Tg' and the crystallization (or devitrification) temperature. DSC measurements show that the residual TBA in the glassy state will cause a decrease in Tg' and will crystallize during heating. An increase in the cooling rate causes a decrease in Tg'. For monophase solution containing different lyoprotectants, the addition of TBA can all cause a decrease of Tg'. The observation of cryomicroscopy shows that the ice crystals are linear. Freeze-drying of monophase solution of TBA/water cosolvent system can be used to prepare proliposomes which forms liposomes after rehydration, and the sizes of liposomes with different lyoprotectants and different concentrations are between 200 and 900 nm.
