Summary

Mature zygotic embryos of Ash were cryopreserved following a simple dehydration treatment. 63% germination was obtained when embryos were dried to a moisture content of 12-14% prior to rapid freezing in liquid nitrogen. Embryos transferred to Murashige and Skoog basal medium for 16-48 hours following excision required additional 0.4 M sucrose preculture to give some protection against freezing damage.
