Summary

A protocol was established for freezing and thawing marmoset spermatozoa which resulted in post-thaw recovery rates of approximately 50% motile sperm. This protocol was used to compare different cooling rates: fast freezing, either in liquid nitrogen vapour or on the surface of a dry ice block, or a slower controlled method using an automated cell freezer. Although no differences were found in length of sperm survival after thawing, some differences in various motility parameters between fast and slow cooling rates were observed using computerised sperm motility analysis.
