Summary

An investigation was carried out as a part of a research on Brucella to improve the quality of complements used in complement fixation test. Two groups of guinea pigs were given supplementary diets consisting of green bean sprouts and ripe papaya fruits for two weeks prior to serum collection, while control guinea pigs were not given any. One day before serum collection the animals were given water only. Serum samples collected were stored at 4, -20, -70 deg C and liquid nitrogen temperature. Research results indicated that the index of working dilution of complement varied. Working dilution of complement indexes of the treated groups were 73, 71, and 65 obtained from guinea pigs from Baturaden, Australia and Bogor, respectively, whilst the figures for the control groups were 14, 16 and 15. This research suggested that complements stored in liquid nitrogen could be maintained for 120 days, 65 days when stored at -70 deg C, 50 days at -20 deg C and 6 days at 4 deg C.
