Summary

The effect of bulb storage temperature on scale explant regeneration was investigated. Bulbs were stored 8 weeks at 4 °C, 22 °C, 30 °C and 40 °C/22 °C (10 hours 40 °C and 14 hours 22 °C). The best results were obtained with treatment of progenitor bulbs at 30 °C. The effect of incubation temperature (15 °C and 22 °C) on shoot explant regeneration was studied. MS medium without growth regulators or supplemented with 1 ppm BA and 0.5 ppm NAA was used. Bulblets were formed more readily at 22 °C and on medium with growth regulators.
