Cryochemistry: peptide coupling in frozen organic solution.

Type of article: Article

Summary

Examples of peptide coupling in frozen dioxane solution are presented and compared to the results obtained in liquid solution. The protected dipeptides of glycine-alanine-, phenylalanine-alanine, -glycine-phenylalanine and phenylalanine-phenylalanine have been synthetized by using of 4-nitrophenyl active esters for the coupling. The reactions carried out at -18 deg C in frozen conditions produced a significantly higher reaction rate than those of 40 deg C. The role of frozen dioxane in the improved results is discussed.
