Cryopreservation of lactate dehydrogenase: interactions among various cryoprotectants.

Summary
Lactate dehydrogenase was exposed to temperatures down to -30 deg C in the presence of inorganic electrolytes and combinations of various cryoprotective solutes. When sugars, polyols and bovine serum albumin were used, cryopreservation was mainly dependent on the concentration ratio of cryoprotectant to enzyme rather than the concentration of inorganic salts in the residual liquid of the frozen system. Since ethylene glycols, methanol and DMSO might favour the detrimental action of inorganic electrolytes during freezing, cryopreservation in the presence of these cryoprotectants seems to be due predominantly to colligative action of the solutes. Conformities in the freeze-thaw behaviour of a globular enzyme and a biomembrane are discussed.
