Summary
THE PRETREATMENT CONDITIONS RELATIVE TO CRYOPROTECTANT CONCENTRATIONS (SUCROSE AND DMSO) WERE STUDIED, IN ORDER TO DEFINE A CRYOPRESERVATION PROCESS FOR IN VITRO CULTURED CARNATION MERISTEMS. MERISTEM RESISTANCE TO DEEP FREEZING IS CLOSELY DEPENDING ON SUCROSE AND, TO A SMALLER EXTENT, ON DMSO CONCENTRATIONS. SOME OPTIMAL CONDITIONS, ENSURING A REGROWTH RATE OF 98.9%, HAVE BEEN DEFINED: PRECULTURE OF EXCISED MERISTEMS, DURING 24 HOURS, ON A MEDIUM SUPPLEMENTED WITH 0.75 M SUCROSE, FOLLOWED BY THE APPLICATION OF A SOLUTION CONTAINING 5% DMSO (V/V). A SLOW FREEZING IS USED: 0.5 K/MIN, DOWN TO 233 K (-40 DEG C), PRIOR TO IMMERSION IN LIQUID NITROGEN, AND THE SAMPLES ARE RAPIDLY REWARMED IN A 313 K (40 DEG C) WATER BATH.
