Summary

THE AUTHORS DESCRIBE 2 DIFFERENT METHODS FOR MEASURING CRYOPROTECTION OF ISOLATED CHLOROPLAST MEMBRANES BY SOLUBLE LEAF PROTEINS. PROTECTION AGAINST MECHANICAL FREEZE-THAW DAMAGE IS QUANTIFIED EITHER IMMUNOCHEMICALLY AS THE LOSS OF THE SOLUBLE LUMENAL (INSIDE) PROTEIN PLASTOCYANIN OR BY MEASUREMENT OF THE THYLAKOID VOLUME CHANGES ASSOCIATED WITH OSMOTIC RUPTURE. BOTH METHODS YIELD COMPARABLE RESULTS. THE IN VITRO ASSAYS ARE USED TO FOLLOW CRYOPROTECTIVE PROTEINS THROUGH DIFFERENT FRACTIONATION STEPS.
