Summary

THE PAPER DESCRIBES THE TECHNIQUE BY WHICH CHLAMYDIA PSITTACI ORGANISMS WERE FROZEN AT LIQUID NITROGEN TEMPERATURE THEN TREATED WITH OSMIUM TETROXIDE, 4%, IN ACETONE AT 194 K (-79 DEG C). COMPARABLE SPECIMENS WERE TREATED WITH OSMIUM TETROXIDE, 1%, AND DEHYDRATED IN ETHANOL. WITH THE FREEZE-SUBSTITUTION FIXATION METHOD THE CYTOPLASMIC MEMBRANE WAS WELL PRESERVED AND SURFACE PROJECTIONS WERE READILY SEEN. THEY WERE NOT SEEN IN THE SPECIMENS TREATED BY THE OTHER METHOD.
