Summary

The mechanisms of damage to a human epidermal model during cryopreservation were examined through the use of a fluorescent multiple endpoint assay. Both plasma membrane integrity and epidermal permeability were susceptible to freezing damage, with reduced damage in the presence of cryoprotective agents. The multiple endpoint assay can be used to assess cryopreservation protocols on the basis of structural integrity.
