Summary

DESCRIPTION OF A TWO-STAGE FREEZING PROCEDURE FOR HUMAN BONE MARROW, OF WHICH A CELL SUSPENSION IS PLACED IN 2-ML POLYPROPYLENE TUBES PLUNGED 20 MIN IN BATHS AT A TEMPERATURE ADJUSTED TO 245, 241, 236 OR 232 K (-28, -32, -37 OR -41 DEG C). THE TUBES ARE THEN DIRECTLY PLUNGED IN LIQUID NITROGEN AND REMAIN THERE 30 MIN. THAWING IS QUICK WATER-BATH AT 315 K (42 DEG C). FOR GRANULOCYTE PRECURSORS, THE OPTIMUM FREEZING TEMPERATURE SEEMS TO BE 236 K. THE TESTS SHOULD BE FURTHERED FOR ERYTHROBLAST PRECURSORS, BUT THEY SEEM TO BE ENCOURAGING. G.G.
