Summary

Cells were cooled to -80 deg C at rates ranging from 0.2 to 10 deg C/minute in the presence of 10% (v/v) cryoprotectant (DMSO, propane-1, 2-diol or glycerol). After freezing and thawing, there was a very strong influence of cooling rate on cell survival in monolayers. Survival was highest (higher than 80%) after cooling at 0.2 deg C/minute, but declined sharply to lower than 40% after cooling at 1 deg C/minute. Conversely, isolated cells showed high survival at 1 deg C/minute but lower survival at 0.2 deg C/minute: indeed cell survival after cooling at 0.2 deg C/minute was lower in suspensions than in monolayers.
