Examination of sugars as excipients for the stabilization of lactate dehydrogenase.

Résumé

Effects of shelf temperature on residual activities of lactate dehydrogenase (LDH) were investigated in view of its practical importance in the ambient temperature storage. Raffinose and trehalose were used as excipients. The mixtures of LDH, phosphate buffer and an excipient were freeze-dried. They were stored at four shelf-temperatures (-25 °C, ambient temperature: 18-25, 50 and 70 °C) for 7-30 days. From the results, raffinose and trehalose maintained LDH activities at higher temperature, 50 °C storage, almost the same as at the ambient temperature storage. Glass transition temperature of the excipients is the most important factor for stabilizing the activity of LDH.
