Summary

CONTINUOUS CRYO-SCANNING ELECTRON MICROSCOPY (SEM) AND FREEZE REPLICA OBSERVATIONS WERE CARRIED OUT ON MOUSE BLASTOCYSTS USING A VITRIFICATION SOLUTION (VS1, 10% GLYCEROL + 20% PROPYLENE GLYCOL AND VS2, 25% GLYCEROL + 25% PROPYLENE GLYCOL). CRYO-SEM OBSERVATION OF BLASTOCYSTS USING A QUICK FREEZ ING SOLUTION (QFS, 3 M GLYCEROL + 0.25 M SUCROSE) WAS ALSO PERFORMED. EXAMINATION OF THE VS REVEALED THE ABSENCE OF BOTH INTRA-AND EXTRACELLULAR ICE CRYSTALS, WHILE THE EXAMINATION OF THE QFS SHOWED THE PRESENCE OF SMALL INTRA-AND EXTRACELLULAR ICE CRYSTALS. THE VITRIFIED BLASTOCYSTS SHOWED MARKED SHRINKAGE AS COMPARED TO THE QUICKLY FROZEN BLASTOCYST.
