EFFECTS OF THE FREEZING PROCESS ON THE SURVIVAL AND REGROWTH RATE OF OIL PALM (ELAEIS GUINEENSIS JACQ) SOMATIC EMBRYOS AFTER STORAGE IN LIQUID NITROGEN.

EFFETS DU MODE DE CONGELATION SUR LA SURVIE ET LA REPRISE DE LA PROLIFERATION D'EMBRYONS SOMATIQUES DE PALMIER A HUILE (ELAEIS GUINEENSIS JACQ) APRES LEUR CONSERVATION DANS L'AZOTE LIQUIDE.

Summary

THE RAPID FREEZING RATE (200 K/MIN) USED INITIALLY FOR THE CRYOPRESERVATION OF OIL PALM SOMATIC EMBRYOS ENSURES RECOVERY RATES OF 8 AND 15% RESPECTIVELY FOR THE 2 CLONES STUDIED. A TWO-STAGE FREEZING METHOD, WHICH COMPRISES A CONTROLLED FREEZING (0.5 TO 40 K/MIN) FROM 293 TO 173 K (20 TO -100 DEG C), FOLLOWED BY THE IMMERSION OF THE SAMPLES IN LIQUID NITROGEN, ENSURES SIMILAR RESULTS WITH FREEZING RATES FASTER THAN 1 K/MIN. THE LARGE EXTENT OF THE USABLE FREEZING RATES SHOULD ALLOW THE DETERMINATION OF A SUITABLE RATE FOR EVERY CLONE. THE USE OF A PROGRAMMABLE FREEZER IS NOT OBLIGATORY FOR THE CRYOPRESERVATION OF SUCH A MATERIAL.
