Summary

SPERM OF SEA URCHINS STRONGYLOCENTROTUS NUDUS AND STRONGYLOCENTROTUS INTERMEDIUS) WAS COOLED TO 77 K IN SEA WATER WITH CRYOPROTECTIVE ADDITIVES (1 M GLYCEROL, 1.5 M DMSO AND 1.5 M ETHYLENE GLYCOL). ICE CRYSTALLIZATION WAS ARTIFICIALLY INITIATED AT 266 K (-7 DEG C). UP TO 233 K (-40 DEG C), THE SPERM WAS COOLED AT A GRADUALLY INCREASING RATE FROM 0.3 TO 1.5 K/MIN. THEN THE SAMPLES WERE TRANSFERRED INTO LIQUID NITROGEN. THAWING WAS PERFORMED IN WATER BATH AT 293 K (20 DEG C). ETHYLENE GLYCOL AND DMSO PROVED TO BE THE BEST CRYOPROTECTANTS.
