Summary

USING BRITISH STANDARD (NO. 4285: 1968) METHODS FOR SAMPLING PLATE PREPARATION AND INOCULATION OF PLATES, PSYCHROTROPHIC COUNTS AFTER INCUBATION AT 294 K (21.C) FOR 25 H CORRELATED WELL (R =+0.97, P LOWER THAN 0.001) WITH THOSE OBTAINED AFTER CONVENTIONAL INCUBATION AT 279 K (6.C) FOR 14 DAYS. THE CORRELATION WAS HIGHER WITH A PREDOMINANTLY GRAM-NEGATIVE THAN WITH A GRAM-POSITIVE CREAM MICROFLORA (R =+0.98 VS. R=+0.91). DISTRIBUTION OF BACTERIAL TYPES WAS SIMILAR FOR BOTH INCUBATION PROCEDURES. WHEN A SPIRAL PLATE MAKER WAS USED FOR SURFACE PLATING OF THE SAMPLES, COUNTS CORRELATED WELL (P LOWER THAN 0.001) WITH CORRESPONDING COUNTS OBTAINED WITH THE STANDARD POUR PLATE TECHNIQUE, BUT WERE HIGHER. IT IS SUGGESTED THAT SOME PSYCHROTROPHS MAYBE UNABLE TO WITHSTAND THE TEMP. OF MOLTEN AGAR USED IN THE POUR PLATE METHOD WHICH WOULD THUS UNDERESTIMATE THE NUMBER OF PSYCHROTROPHS PRESENT.
