Summary

A method is described to calculate cell volume during exposure to and removal of cryoprotective compounds. The osmolality of the solutions is calculated by summing the values for individual components on a molal basis and applying experimentally-determined or assumed osmotic coefficients. Transient volumes are calculated assuming only osmotically-driven water flux, and equilibrium volumes are calculated assuming complete penetration of low-molecular weight cryoprotectants. The calculations are used to design procedures to reduce osmotic injury during corneal cryopreservation.
