Summary

THE LONG TERM DEXTER CULTURE SYSTEM ALLOWS FOR THE STUDY OF HEMOPOIETIC AND STROMAL ELEMENTS. THE AUTHORS HAVE USED THIS SYSTEM TO EVALUATE THE CRYOPRESERVATION OF BOTH HEMOPOIETIC ANS STROMAL CELLS. C57BL MURINE MARROW WAS ASSAYED FOR STEM CELLS AND GROWN IN DEXTER CULTURE BOTH BEFORE AND AFTER CRYOPRESERVATION USING CONTROLLED MODE COOLING IN A PROGRAMMABLE UNIT AND STORAGE AT 118 K (-155 DEG C). MARROW CELLS COOLED AT TWO DIFFERENT RATES TO THE EUTECTIC POINT 1K/MIN TO 243 K (-30 DEG C) AND 7K/MIN TO 243K WERE EVALUATED. THE DATA SHOW THAT BOTH STROMAL AND HEMOPOIETIC STEM CELLS CAN BE EFFECTIVELY CRYOPRESERVED AND THAT THE INCREASED RATE OF FREEZING FACILITATES BOTH STEM CELL AND STROMAL CELL RECOVERY AFTER CRYOPRESERVATION.
