Summary
In the field of clinical organ transplantation, the first step of kidney cryopreservation is filling the isolated kidney with about 4°C cryoprotective agents (CPA). The morphology of functional cells was greatly influenced by cooling rate. When the CPA filled in it is important to study the cell damage induced by cooling rate. The isolated pig kidney was chosen as research object. The samples were separately cooled at a rate of 36, 12, 3.6 and 1.2°C/min from body temperature to 3 and -1°C. After cooling, the kidney sample were observed under optical microscope after fixing, embedding, sectioning and straining. The results shows lower cooling rate has little effects on microstructure damage while higher cooling rate was disadvantageous to cryopreservation by likely causing renal tubular ectasia, necrosis and detachment. The result is useful for clarifying the mechanism of activity of organs.
