Flow cytometric analysis of plasma membrane damages of rainbow trout and turbot frozen spermatozoa.

Summary

The aim of the study was to understand the difference in freeze-thawing survival between trout and turbot spermatozoa by evaluating membrane damage of spermatozoa. Flow cytometric analyses were performed on cryopreserved turbot and trout spermatozoa after fluorescent staining with propidium iodide to assess plasma membrane integrity. The composition of the freezing medium used is given. These penetrating cryoprotectants were tested. The freezing rate was -4/minute until -80 deg C, followed by storage at -196 deg C and thawing to 4 deg C. The results, suggesting different cryoprotectant-membrane interactions in trout and turbot, are discussed in light of membrane composition of spermatozoa of each species.
