ISOTHERMAL NUCLEATION STAGE AND ITS APPLICATIONS IN THE FREEZING OF BIOLOGICAL CELLS.

Summary

THE AUTHORS CONSIDER THE USE OF A STAGE SUBJECTING THE CELLS TO SUBZERO TEMPERATURES, CONSTANT IN SPACE AND TIME. THE COOLING WINDOW IS A SAPPHIRE OF OPTICAL QUALITY, SELECTED FOR ITS HIGH THERMAL CONDUCTIVITY. THE WINDOW TEMPERATURE IS CONTROLLED BY HEATING OR COOLING THE HEAT TRANSFER FLUID SO THAT THE TEMPERATURE IS REACHED. THIS DEVICE DETERMINES THE PROBABILITY OF THE FORMATION OF INTRACELLULAR ICE CRYSTALS OR PREDICTS THE WATER PERMEABILITY OF CELLS AND EMBRYOS. THE RESULTS SHOW ACCEPTABLE VARIATIONS IN THE SPACE DISTRIBUTION OF TEMPERATURE (20 K/M) AND THAT THE TIME VARIATIONS OF TEMPERATURE CAN BE ELIMINATED TO A LARGE EXTENT. THEAUTHORS GIVE THE RESULTS OBTAINED FOR THE TEMPERATURES OF HUMAN B-LYMPHOCYTES AND URCHIN EGGS.
