Summary

THE CULTURE WHITTEN'S MEDIUM WITH HEPES BUFFER HAS BEEN USED AT SUCH STAGES OF CRYOPRESERVATION AS ISOLATION, FREEZING, CULTIVATION, AND TRANSPLANTATION. AFTER THAWING FROM 77 K (-196 DEG C), 80% OF 2-CELL EMBRYOS OF MICE F1 HAD NORMAL MORPHOLOGY, 60% OF NORMAL EMBRYOS DEVELOPED IN VITRO FROM 2-CELL STAGE TO BLASTOCYSTS, AND 36% OF EMBRYOS DEVELOPED INTO NORMAL MICE IF TRANSPLANTED INTO PSEUDOPREGNANT FOSTER MOTHERS. EMBRYO DAMAGES CAUSED BY FREEZING-THAWING AND DMSO EFFECTS WERE PRONOUNCED IN THE PREIMPLANTATION PERIODS OF DEVELOPMENT.
