Summary

THE FREEZE-DRYING OF HAEMOGLOBIN, UNLESS PROTECTED, LEADS TO OXIDATION OF IRON AND FORMATION OF METHAEMOGLOBIN. IT HAS BEEN FOUND THAT THE ADDITION OF TRIS (2-AMINO-2-HYDROXY METHYL PROPANE-1,3-DIOL) TO THE SOLUTION PREVENTS THIS. FURTHER STUDY INTO THE MECHANISM OF PROTECTION IS REPORTED. THE RESULTS OF TRIALS WITH 28 DIFFERENT COMPOUNDS SHOW LEVELS OF METHAEMOGLOBIN AND OXYHAEMOGLOBIN AFTER DESICCATION BY FREEZING TO 233K (-40C) FOLLOWED BY PRIMARY DESICCATION FOR 16 HOURS UP TO 263 K (-10C) AND SECONDARY DESICCATION FOR 8 HOURS UP TO 278 K (5C). DETAILS OF MATERIAL AND METHODS GIVEN. FOUR AMINES WERE STRONGLY PROTECTIVE, NAMELY MONOTRIS (7 ON LIST), BIS-TRIS (8), BIS-TRIS PROPANE (10) AND BIS-AMP (11). FOUR WERE MODERATELY EFFECTIVE, THE REST INEFFECTIVE. THE FUNCTIONS OF THE DIFFERENT GROUPS ARE DISCUSSED. C.R.F.
