Summary

CRYOPRESERVATION OF BOVINE EMBRYOS IS DESCRIBED IN THE PAPER. ON THE WHOLE, 65 EMBRYOS WERE SUBJECTED TO SINGLE-TRACK DEHYDRATION IN 1.5 M GLYCEROL IN PBS (PHOSPHATE-BUFFERED-SALINE), WITH AN ADDITION OF 20% FCS (FETAL CALF SERUM). THE EMBRYOS WERE EXPOSED DIRECTLY TO THE SEEDING TEMPERATURE OF 266 K (-7 DEG C) AND FROZEN STEP BY STEP AT THE RATE OF 0.3-0.5 K/MIN UNTIL 238 K (-35 DEG C) ; THEN THE EMBRYOS WERE TRANSFERRED INTO LIQUID NITROGEN. FOR THAWING, THE EMBRYOS WERE DIPPED IN A WATER BATH AT 308 K (35 DEG C), AFTER WHICH THE CRYOPROTECTIVE AGENT WAS WASHED FROM THEM WITH 1.1 M UE THE RATE OF SURVIVAL WAS 70.8%.
