IN ORDER TO ESTABLISH THE METHOD FOR A LONG-TERM PRESERVATION BY DRYING, FOUR SELECTED STRAINS WERE SUBJECTED TO SEVERAL DRYING AND RECOVERY CONDITIONS USING TWO TYPES OF SUSPENDING MEDIA: SUCROSE + SERUM + SODIUM CHLORIDE AND SODIUM-GLUTAMATE + PHOSPHATE BUFFER + SODIUM CHLORIDE. FREEZE-DRYING GAVE HIGHER VIABILITY DEPENDED UPON THE COMBINATION OF STRAIN AND SUSPENDING MEDIUM. THE ADDITION OF ADONITOL AND CYSTEINE INTO THE SUSPENDING MEDIA PREVENTED DNA STRANDS FROM BREAKING AND IMPROVED VIABILITY. PRESENCE OF MAGNESIUM IONS IN REHYDRATION FLUID INCREASED THE NUMBER OF COLONIES. HYDROGEN PEROXIDE DECOMPOSERS, SUCH AS IRON IONS, SODIUM-PYRUVATE, CYSTEIN OR HAEMOGLOBIN, INCREASED THE NUMBER OF COLONIES WHEN ADDED INTO THE RECOVERY AGAR MEDIA.
