Summary

TWO PLANT CELL SUSPENSION LINES (CATHARANTHUS ROSEUS AND TABAERNAEMONTANA DIVARICATA LINE 21) WERE RECOVERED FOLLOWING FREEZING TO 77 K (-196 DEG C) AT ACCEPTABLE RATE TWO OTHER CELL LINES (CINCHONA ROBUSTA AND TABAERNAEMONTANA DIVARICATA LINE 6) WERE RECALCITRANT TO THESE SAME PROCEDURES. A CRYOMICROSCOPE STUDY INDICATED THAT THE RECALCITRANT LINES SUFFERED A RANGE OF LESIONS DURING FREEZING AND THAWING. A SIGNIFICANT PROPORTION OF THE CELLS OF RESISTANT LINES DEMONSTRATED NONE OF THESE LESIONS WHEN FROZEN IN SUCROSE/DMSO/GLYCEROL (SDG) CRYOPROTECTANT MIXTURES.
