Summary
After crystallization of the suspending medium, cryoprotected protoplasts and cell protoplasm were surrounded by a physically different medium (solid or liquid), yet they showed similar responses to stresses imposed by the freeze-thaw cycle. During freezing, the osmotic effects on both protoplasts and plasmolysed cells were characterized by a marked volumetric shrinkage with considerable distortions of the plasma membrane and subsequent progressive swelling during rewarming. The observations are related to modifications of the plasma membrane and to the cell wall.
