FREEZE-DRYING OF BIOLOGICAL PRODUCTS: FACTS AND PROSPECTS.
Author(s) : SIMATOS D., BLOND G., LE MESTE M.
Summary
THE MOST REMARKABLE PROPERTIES OF LYOPHILIZED PRODUCTS ARE: THE POSSIBILITY OF ATTAINING A VERY LOW RESIDUAL MOISTURE LEVEL AT RELATIVELY LOW TEMPERATURES ; REIMBIBITION OF LIQUID BY THE FREEZE-DRIED MATERIAL ; THE POSSIBILITY OF DRYING MATERIALS HAVING A CELLULAR OR OTHER FINE STRUCTURAL ORGANIZATION. THE FREEZE-DRYING PROCESS HAS FOUND APPLICATIONS IN EXTREMELY VARIOUS FIELDS (PRESERVATION OF EXPERIMENTAL THERMOLABILE MATERIALS, PREPARATORY METHOD OF SAMPLES TO BE OBSERVED BY OPTIC OR ELECTRONIC MICROSCOPY). THE PHARMACEUTICAL AND BIOLOGICAL INDUSTRIES MAKE USE OF FREEZE-DRYING (PLASMA AND OTHER BLOOD FRACTIONS, TISSUE EXTRACTS, ANTIBIOTICS, TEST REAGENTS, MICROORGANISM STRAINS, VACCINES, ETC). EXTENSIVE RESEARCH HAS ANALYSED THE INFLUENCE OF THE PROCESSING CONDITIONS ON THE HEAT AND VAPOUR TRANSFER PHENOMENA DURING THE DRYING PROCESS. THE KNOWLEDGE THUS OBTAINED HAS RESULTED IN IMPORTANT PROGRESS. FINALLY, THE S TO BE USED IN ORDER TO ATTAIN A DETERMINED PROPERTY, ARE STRONGLY DEPENDENT ON THE MATERIAL CHARACTERISTICS. IN PRACTICE TO OBTAIN SATISFACTORY RESULTS IT IS NECESSARY TO USE CRYOPROTECTIVE ADDITIVES, THE USE OF WHICH, STILL LARGELY EMPIRICAL, SHOULD BECOME MORE RATIONAL.
