Summary

STUDIES ON BACTERIAL L-FORMS FREQUENTLY INVOLVE SERIAL PROPAGATION AND MAINTENANCE OF CULTURES WHEN NOT IN REGULAR USE IS A PROBLEM OF SOME MAGNITUDE. THE STUDY WAS UNDERTAKEN TO OBTAIN INFORMATION ON THE EFFECT OF STORAGE AT LOW FREEZING TEMPERATURES ON SOME BACTERIAL L-FORMS. FOUR STRAINS OF STAPHYLOCOCCUS AUREAS L-FORMS, STREPTOCOCCUS PYOGENES L-FORMS TWO STRAINS, AND A STRAIN OF ESCHERICHIA COLI WERE USED. THESE HAD BEEN SUB-CULTURED TWICE WEEKLY DURING FOUR YEARS. CULTURES IN BHI (BRAIN HEART INFUSION) BROTH + 5% NACL + 10% HORSE SERUM WITH A CONCENTRATION OF ABOUT 108 COLONY FORMING UNITS PER MILLILITRE WERE PLACED IN TEST TUBES (3 ML PER TUBE) AND HELD AT ROOM TEMPERATURE, 4 DEG C, -20 DEG C AND -80 DEG C. TABLES SHOW THE EFFECTS OF FREEZING 1. ON COLONY FORMING UNITS, 2. ON SURVIVAL OF CFU AFTER THREE YEARS STORAGE. SURVIVAL WAS HIGHEST AT -80 DEG C, 3. THE EFFECTS OF ADDING THE ANTI-FREEZES GLYCEROL AND DIMETHYL R. F.
