Vitrification of Oryza sativa L. cell suspensions.

Summary

Cells were loaded with a 1.5 M DMSO solution at 20 deg C for 20 minutes, and then dehydrated with a vitrification solution of 44.5 wt% DMSO + 18.7 wt% sorbitol at 0 deg C for 4 minutes. After quenching in liquid nitrogen, the cells were warmed and unloaded in a 0.5 M sorbitol solution at 20 deg C for 30 minutes. A few colonies were formed during the post-thaw culture. The cell suspension resulting from the colonies grew in the same manner as the original cell suspension.
