Summary

The effect of the cooling rate on intracellular ice formation was investigated between 20 and 400 deg C/min in isotonic solution. The temperature at which 50% of hepatocytes showed intracellular ice formation was almost constant with an average value of -7.7 deg C. Different behaviour was seen in the presence of extracellular ice. These experiments clearly demonstrated both the time and temperature dependence of intracellular ice formation. On the other hand, in cooling experiments in the absence of extracellular ice, intracellular ice formation was not observed until about -20 deg C, suggesting the involvement of the external ice in the initiation of intracellular ice formation. The effect of DMSO on intracellular ice formation was also quantified. The results suggest that multistep protocols will be required for the cryopreservation of hepatocytes.
