Summary
The paper describes methods of platelet concentrate (PC) freezing and results of platelet aggregation of thawed PC. To get rid of the bad smell of dimethylsulfoxide and the washing procedure of thawed PC before transfusion, the author chose glycerol, as the cryoprotectant. Three freezing methods in the presence of 5 percent glycerol were done. After 9 years storage at less than -150 deg C, thawed PC frozen by the sample control method is aggregated at almost prefreeze levels. By the chamber control method, thawed PC is aggregated at the level of less than that of sample control. A program freezer is necessary for the freezing of such delicate cells. A single sample of PC divided into several aliquots was frozen successfully by the sample control method. Many different PC solutions were frozen by the chamber control with less aggregation. Further studies are needed for setting the optimum freezing conditions for the latter purpose.
