Summary
THE EFFECT OF DMSO AND HES ON VIABILITY AND FUNCTIONAL CAPACITY OF HUMAN GRANULOCYTES WERE ASSESSED AFTER INCUBATION OF GRANULOCYTES WITH CRYOADDITIVES AT 277 K (4 DEG C). RESULTS SHOWED THAT A CONCENTRATION OF DMSO HIGHER THAN 6% WAS TOXIC FOR GRANULOCYTES AFTER AN INCUBATION TIME OF 15 MINUTES, WHILE HES EXHIBITED NO TOXICITY AT CONCENTRATIONS AS HIGH AS 15%. HUMAN GRANULOCYTES CRYOPRESERVED WITH 5% DMSO AND 5% HES SHOWED A GOOD RECOVERY OF VIABLE CELLS, BUT ON THE CONTRARY A SIGNIFICANT IMPAIRMENT OF THEIR FUNCTIONAL CAPACITY. IN PARTICULAR, FROZEN-THAWED GRANULOCYTES QUICKLY LOST THEIR VIABILITY WHEN THEY WERE WASHED AND STORED AT 310 K (37 DEG C).
