Freezing induced protein changes in fish evaluated by two-dimensional gel electrophoresis.

Summary

An approach has been established to apply the technique of two-dimensional gel electrophoresis (2-DE) to monitor freezing induced changes in muscle proteins from cod (Gadus morhua). In each gel an average of 435 more or less 61 spots were found. Comparing the two-dimensional (2-D) protein patterns reveals 150 spots representing proteins which might be sensitive to freezing. The spots either disappear from or appear in the 2-D pattern from muscle which has been frozen as compared to the 2-D pattern from fresh muscle. Differences between 2-D patterns from muscles which have been frozen stored under different conditions were also found. Based on 2-D protein patterns a model to discriminate between fresh and frozen cod and between cods frozen stored under different conditions was built using multivariate analysis.
